DAB (3,3'-diaminobenzidine hydrochloride, MW = 214.1) reacts with HRP in the presence of peroxides to produce an insoluble brown product at the site where the peroxidase-coupled antibody binds to the sample. The brown precipitate is insoluble in alcohol and other organic solvents, making it an excellent substrate for immunohistochemical staining that requires the use of conventional counterstaining and blocking agents.The Pierce DAB Substrate Kit includes a 10X DAB solution and a stabilized peroxide substrate buffer. Crystallized DAB (powder) is also available.
